You should specify that we're talking about main characters, Gravity has a few characters in the beginning, and My Dinner With Andre has the waiter.

ElM. some of those actually have one or two onscreen characters, but movies which have a few brief figures and/or voices apart from that should definitely count anyway.

"Two main characters" seems too loose. "Only two significant characters" may be better.

if we are talking 'two main characters', then we have to include my favorite film of all time...'chinese coffee' starring jerry orbach and al pacino...

a brilliant movie that started out as a two man play...(the movie just throws in a few quick unnecessary flashbacks)...

Ooh, I LOVE minimalist movies like that, so behold my pile of suggestions ranging from no budget efforts to real masterpieces of craft. I'd love to a few movies like that myself, but making them right actually require professionalism and experience. Ironically enough, the less is going on the harder is to make the movie.  

FYC:
Un homme qui dort (1974) (one character not counting the narrator; an AMAZING movie) 
Secret Honor (1984) (one character, Richard Nixon)
The Caller (1987) (two characters; virtually unknown cult masterpiece, a great suspenseful thriller with one of the best endings ever)
All Is Lost (2013) (one character) 
Tayna Slendermena (2013) (two characters)
Kollektor (2016) (one character, very similar to Locke)
The Last Days of Earth (2017) (okay, I am actually not sure whether this San-Marino/Ukraine no budget co-production starring yours truly counts ... All of the discernible characters were portrayed by me, but I'm not entirely sure whether I was portraying three different people or all of them are one. I guess that is left to interpretation)
Cursed Forest (2018) (one and a half character)
Chaos (2019) (two characters; one of them is portrayed by me)

Kyle Perez only my voice, though, but yes. In 2008 it was a bit of a dream to be an extra of some sort in a Batman movie and in 2012 it suddenly came through. :) I had a sudden repeat of the situation in 2018 as my voice is also technically in Bohemian Rhapsody (2018), among tens of thousands on Live Aid. I guess that would be more of a miscellaneous crew credit then an actor one.  

Dan Dassow, thanks! Most of those are actually no/low budget efforts including my own productions, as well as quite a few moments for which I'm not proud of (if not ashamed). Then again, experience gained is valuable anyway. :)

Hoekkie interestingly, I thought about Cast Away (2000) but dismissed it because I remember a lot of minor characters from it. 

Still, for the most of the movie it fits well into the poll, i guess. I mean, there a reason why volleyball is a famous character here. :)

My favorite from this list is Sleuth.

I have only seen a few scenes from My Dinner with Andre, but I suspect when I watch the whole film, it'll be one of my all-time favorites.

Nikolay, I had recorded The Caller from TV (ahh ,the good old Betamax days) after reading about it in Fangoria magazine. It definitely has a crazy ending, but the movie is really very suspenseful. So far you're the only person I know that actually mentioned it.

And I'm very surprised nobody has yet mentioned Hell in the Pacific (1968). (Sorry if someone has mentioned and I missed it)
